Differential Expression of Vimentin and GFAP Protein during Brain Development of Mouse Fetuses after Treated with 2-Methoxyethanol

Abstract. This study aims to determine the differences on protein expression of vimentin and GFAP in the brains of fetal mice Mus musculus (Swiss Webster) during the period of brain development ( at the GD-11 to the GD-18), caused by 2-methoxyethanol (2ME) that injected intraperitoneally with a dose of 7,5 mmol/kg body weight at gestation days (GD) 10. In the last decade, it has been widely reported the negative effects of chemicals polluting the environment. 2-methoxyethanol is one of the chemical pollutants, which is teratogenic, embryotoxic or teratogenic in both animals and humans. 2-ME and its metabolites in the body can damage cells and tissues. The body can be exposed by 2-ME through the air, water and soil. Fetal brains were isolated and collected, into the tube containing a solution of Cell Lytic M reagent. Protein concentration was measured by BCA nanodrops 2000 and the Smart Kit Cat No. 21071 Japan. To determine the protein expression immunodot blot was performed by using primary antibodies vimentin and GFAP. The amount of protein expressed was measured by immnodotblot and densitometry techniques.. The results showed that the expression of vimentin was always higher than GFAP in the brains of fetal at GD-11 until GD-18. The highest expression of vimentin was highest in fetal brain at GD-17 and18 days. The highest expression of GFAP present in fetal brain GD-12. Generally, it could be concluded that the protein Vimentin is very necessary during the period of fetal brain development, especially for healing wounds caused by 2-Methoxyethanol.
